Wechselspiel von Zelladhäsion und Matrixproteolyse in barrierebrechenden Invadopodien von Tumorzellen

    Adhäsion an die extrazelluläre Matrix und deren Proteolyse durch Matrixmetalloproteinasen (MMPs) sind für die Metastasierung wesentlich. Fibrilläres Kollagen-I und vermutlich neu synthetisiertes VE-Cadherin fördern auf Krebszellen die Expression von MMP14, die mit einer FRET-Sonde messbar ist. Die MMP14-Stimulation durch supramolekulare Orientierung von Kollagenfibrillen und durch den krebszelltypischen Cadherin-Schalter werden untersucht. Translational wird das Potential von MMP14, die krebsselektive Endozytose von Arzneimitteln in metastatischen und gefäßintegrierten Krebszellen zu aktivieren, untersucht.
