10. Simultaneous targeting of tumour cells and stroma by pH-sensitive engineered biologicals

The aim of the project is the generation of efficacious and convenient therapy agents using a multi-level strategy. Tumor cells will be eliminated by a combination of cytotoxic agents with immunomodulators aimed to compensate for the poor immunological response against PDAC. This aggressive approach requires a tightly controlled targeting and delivery to avoid undesired effects; specific targeting to the tumour will be achieved by directing the agent towards ion channels and transporters particularly enriched in the surface of tumour cells. Once the therapeutic agent is accumulated in the tumor, release of the active components will be triggered by the physico-chemical characteristics of the tumour microenvironment, in order to reduce the incidence of undesired effects.
